Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004172.5(SLC1A3):c.922G>A (p.Gly308Ser), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with SLC1A3-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with serine at codon 308 of the SLC1A3 protein (p.Gly308Ser). The glycine residue is highly conserved and there is a small physicochemical difference between glycine and serine.
