This variant lies in the SEC63 gene (transcript NM_007214.5) at coding-DNA position 2006 through coding-DNA position 2007, deleting 2 bases; at the protein level this means shifts the reading frame starting at histidine residue 669, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The SEC63 c.2006_2007delAT variant is predicted to result in a frameshift and premature protein termination (p.His669Argfs*41). This variant was reported in a family with autosomal dominant polycystic liver disease and co-segregated with disease in five affected family members (Table 1 and Supplemental Figure 1, Davila S et al 2004. PubMed ID: 15133510). This variant has not been reported in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare.
