This variant lies in the PLEC gene (transcript NM_201378.4) at coding-DNA position 66, where C is replaced by G; at the protein level this means converts the codon for tyrosine at residue 22 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing. ClinVar contains an entry for this variant (Variation ID: 1251951). This variant has not been reported in the literature in individuals affected with PLEC-related conditions.
