Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001365276.2(TNXB):c.8317A>T (p.Arg2773Trp), citing Ambry Variant Classification Scheme 2023: The p.R2773W variant (also known as c.8317A>T), located in coding exon 23 of the TNXB gene, results from an A to T substitution at nucleotide position 8317. The arginine at codon 2773 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
