Uncertain significance — the classification assigned by Ambry Genetics to NM_006506.5(RASA2):c.1116T>A (p.Asp372Glu), citing Ambry Variant Classification Scheme 2023: The p.D372E variant (also known as c.1116T>A), located in coding exon 11 of the RASA2 gene, results from a T to A substitution at nucleotide position 1116. The aspartic acid at codon 372 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
