This variant lies in the MAP2K1 gene (transcript NM_002755.4) at coding-DNA position 901, where G is replaced by A; at the protein level this means replaces glycine at residue 301 with arginine — a missense variant. Submitter rationale: Variant summary: MAP2K1 c.901G>A (p.Gly301Arg) results in a non-conservative amino acid change located in the Protein kinase domain (IPR000719) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251480 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.901G>A in individuals affected with Cardiofaciocutaneous Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
