Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.5281T>C (p.Tyr1761His), citing Ambry Variant Classification Scheme 2023: The p.Y1753H variant (also known as c.5257T>C), located in coding exon 30 of the FLNA gene, results from a T to C substitution at nucleotide position 5257. The tyrosine at codon 1753 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
