This variant lies in the LAMB2 gene (transcript NM_002292.4) at 5 bases into the intron immediately after coding-DNA position 4573, where G is replaced by A. Submitter rationale: This sequence change falls in intron 27 of the LAMB2 gene. It does not directly change the encoded amino acid sequence of the LAMB2 protein. It affects a nucleotide within the consensus splice site. This variant is present in population databases (rs372438959, gnomAD 0.02%). This variant has been observed in individual(s) with clinical features of LAMB2-related conditions (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1210226). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
