NM_005633.4(SOS1):c.2684G>A (p.Ser895Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S895N variant (also known as c.2684G>A), located in coding exon 17 of the SOS1 gene, results from a G to A substitution at nucleotide position 2684. The serine at codon 895 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
