Submitter rationale: The p.Y68* pathogenic mutation (also known as c.204C>A), located in coding exon 3 of the PTEN gene, results from a C to A substitution at nucleotide position 204. This changes the amino acid from a tyrosine to a stop codon within coding exon 3. This alteration has been reported in individuals meeting relaxed International Cowden Consortium operational criteria for Cowden syndrome and in an individual with Bannayan-Riley-Ruvalcaba syndrome (Sarquis MS et al.
