NM_001365276.2(TNXB):c.9725G>A (p.Arg3242His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R3240H variant (also known as c.9719G>A), located in coding exon 27 of the TNXB gene, results from a G to A substitution at nucleotide position 9719. The arginine at codon 3240 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
