Likely pathogenic for Autosomal recessive limb-girdle muscular dystrophy — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001130987.2(DYSF):c.4794dup (p.Ile1599fs), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the DYSF gene (transcript NM_001130987.2) at coding-DNA position 4794, duplicating one base; at the protein level this means shifts the reading frame starting at isoleucine residue 1599, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Variant summary: DYSF c.4677dupC (p.Ile1560HisfsX41) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been reported in affected individuals (HGMD). The variant was absent in 251462 control chromosomes (gnomAD). To our knowledge, no occurrence of c.4677dupC in individuals affected with Limb-Girdle Muscular Dystrophy, Autosomal Recessive and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
