Pathogenic for Postlingual sensorineural hearing impairment; Progressive sensorineural hearing impairment; Bilateral sensorineural hearing impairment — the classification assigned by Laboratory of Human Genetics, Institute of Biosciences - University of Sao Paulo to NM_181659.3(NCOA3):c.2810C>G (p.Ser937Cys), citing ClinGen HL ACMG Specifications v1. This variant lies in the NCOA3 gene (transcript NM_181659.3) at coding-DNA position 2810, where C is replaced by G; at the protein level this means replaces serine at residue 937 with cysteine — a missense variant. Submitter rationale: pathogenic missense heterozygous variant was found to segregate with HL in ten members of the same family, two members had the variant but they did not manifest the HL to date. Neverthless, functional sutdies demostrated that a knockout zebrafish has altered development of the inner ear and the adult spercimes exhibit abnormal hearing behaviour
