This variant lies in the WDR45 gene (transcript NM_001029896.2) at coding-DNA position 755, where T is replaced by C; at the protein level this means replaces leucine at residue 252 with proline — a missense variant. Submitter rationale: A missense variant, c.755T>C in exon 9 of WDR45 is observed in heterozygous state in proband. On segregation, the variant was present in heterozygous state in proband and is absent in her mother and brother (Lab ID: 26138). Father sample was not available for testing. This variant is absent in the gnomAD (v4.1.0) population database and in our in-house database of 4303 exomes. This variant is reported in ClinVar as likely pathogenic by a single submitter (Accession: VCV001184258.3). In-silico analysis tools (CADD_phred and MutationTaster) predict the variant to be disease-causing and likely to affect the WDR45 protein function.
