NM_201253.3(CRB1):c.548G>A (p.Cys183Tyr) was classified as Likely pathogenic for Retinal dystrophy by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: CRB1 c.548G>A (p.Cys183Tyr) results in a non-conservative amino acid change located in the EGF-like domain (IPR000742) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251390 control chromosomes. c.548G>A has been reported in the literature in homozygous individuals affected with Retinitis Pigmentosa and/or Leber congenital amaurosis (e.g. Abolhassani_2024, Tayebi_2019). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 38374194, 30820146). ClinVar contains an entry for this variant (Variation ID: 1180656). Based on the evidence outlined above, the variant was classified as likely pathogenic.
