Likely pathogenic for Mitochondrial DNA depletion syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004614.5(TK2):c.311G>A (p.Arg104His), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TK2 c.311G>A (p.Arg104His) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 8e-06 in 250982 control chromosomes (gnomAD). c.311G>A has been observed in individuals affected with features of Mitochondrial DNA Depletion Syndrome - TK2 Related (Garone_2018, Abolhassani_2024). These data indicate that the variant may be associated with disease. A different variant affecting the same codon has been classified as likely pathogenic/pathogenic by our lab (c.310C>T, p.Arg104Cys), supporting the critical relevance of codon 104 to TK2 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
