This variant lies in the GBA1 gene (transcript NM_000157.4) at 12 bases into the intron immediately before coding-DNA position 1506, where C is replaced by T. Submitter rationale: Variant summary: GBA c.1506-12C>T alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00082 in 248526 control chromosomes in the gnomAD database, including 2 homozygotes. This frequency is not significantly higher than expected for a pathogenic variant in GBA causing Gaucher Disease (0.00082 vs 0.005), allowing no conclusion about variant significance.
