NM_000052.7(ATP7A):c.1592G>T (p.Arg531Met) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R531M variant (also known as c.1592G>T), located in coding exon 5 of the ATP7A gene, results from a G to T substitution at nucleotide position 1592. The arginine at codon 531 is replaced by methionine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
