Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000400.4(ERCC2):c.720C>G (p.Asp240Glu), citing Ambry Variant Classification Scheme 2023: The p.D240E variant (also known as c.720C>G), located in coding exon 9 of the ERCC2 gene, results from a C to G substitution at nucleotide position 720. The aspartic acid at codon 240 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
