This variant lies in the ERCC2 gene (transcript NM_000400.4) at coding-DNA position 733, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 245 with asparagine — a missense variant. Submitter rationale: Variant summary: ERCC2 c.733G>A (p.Asp245Asn) results in a conservative amino acid change located in the DEAD2 domain (IPR010614) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.2e-05 in 250228 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.733G>A in individuals affected with Xeroderma Pigmentosum and no experimental evidence demonstrating its impact on protein function have been reported.
