This variant lies in the ERCC6L2 gene (transcript NM_020207.7) at coding-DNA position 1973, where G is replaced by A; at the protein level this means replaces serine at residue 658 with asparagine — a missense variant. Submitter rationale: This homozygous missense variant of ERCC6L2 was identified in a 12 year old male with pancytopenia and a hypocellular bone marrow who had failure to thrive, thin teeth and muscle pain (29987015, family 2). His youngers sister, who is also homozygous for this variant, was found to have a hypocellular bone marrow and a raised HbF. The following ACMG/AMP criteria were used: PM2, PP3 and PP1.
