This variant lies in the STAT3 gene (transcript NM_139276.3) at coding-DNA position 1229, where A is replaced by G; at the protein level this means replaces histidine at residue 410 with arginine — a missense variant. Submitter rationale: Variant summary: STAT3 c.1229A>G (p.His410Arg) results in a non-conservative amino acid change located in the DNA-binding domain (IPR013801) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251446 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrences of c.1229A>G have been reported in individuals affected with Hyper IgE Syndrome. However, somatic occurrences of this variant have been reported in individuals affected with various hematopoietic and lymphoid malignancies (Example: Andersson_2016, Song_2018). In addition, the variant was also reported an individual affected with triple-negative breast cancer (TNBC) (Young-Shin_2018). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance for Hyper IgE Syndrome until additional information becomes available to determine its role in the Hyper IgE syndrome.
