The affected amino acids are in the ETS domain where damaging ETV6 variants are known to cluster (PM1; PMID: 32693409, 26522332). To our knowledge, functional studies affecting these amino acids have not been performed. This variant has been reported in an individual with thrombocytopenia and B cell acute lymphoblastic leukemia (PS4_Supporting; internal data). In summary, this variant meets criteria to be classified as of uncertain significance based on the ACMG/AMP criteria: PM1, PM2_Supporting, PS4_Supporting.
