NM_000090.4(COL3A1):c.1387G>A (p.Glu463Lys) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E463K variant (also known as c.1387G>A), located in coding exon 20 of the COL3A1 gene, results from a G to A substitution at nucleotide position 1387. The glutamic acid at codon 463 is replaced by lysine, an amino acid with similar properties. This variant was reported in individual(s) with features consistent with Ehlers-Danlos syndrome (Wei X et al. Front Genet, 2023 Dec;14:1246712). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
