Uncertain significance for Familial hypercholesterolemia — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_174936.4(PCSK9):c.1696T>C (p.Trp566Arg), citing ACMG Guidelines, 2015: This missense variant replaces tryptophan with arginine at codon 566 of the PCSK9 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two individuals affected with familial hypercholesterolemia as well as in five healthy control individuals (PMID: 33147992). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
