Uncertain significance for Cardiomyopathy — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_005159.5(ACTC1):c.809-1G>A, citing ACMG Guidelines, 2015: This variant causes a G to A nucleotide substitution at the -1 position of intron 5 of the ACTC1 gene. Splice prediction tools suggest that this variant may disrupt RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 1/246456 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Clinical relevance of loss-of-function truncation and splice variants in the ACTC1 gene is not clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
