NM_000059.4(BRCA2):c.3360A>C (p.Glu1120Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1120D variant (also known as c.3360A>C), located in coding exon 10 of the BRCA2 gene, results from an A to C substitution at nucleotide position 3360. The glutamic acid at codon 1120 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
