Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002485.5(NBN):c.28C>T (p.Pro10Ser), citing Ambry Variant Classification Scheme 2023: The p.P10S variant (also known as c.28C>T), located in coding exon 1 of the NBN gene, results from a C to T substitution at nucleotide position 28. The proline at codon 10 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
