Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.2518G>C (p.Asp840His), citing Ambry Variant Classification Scheme 2023: The p.D840H variant (also known as c.2518G>C), located in coding exon 16 of the ATM gene, results from a G to C substitution at nucleotide position 2518. The aspartic acid at codon 840 is replaced by histidine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
