Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.503A>G (p.Asn168Ser), citing Ambry Variant Classification Scheme 2023: The p.N168S variant (also known as c.503A>G), located in coding exon 6 of the MLH1 gene, results from an A to G substitution at nucleotide position 503. The asparagine at codon 168 is replaced by serine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. In addition, this alteration is predicted to be benign by MAPP-MMR in silico analyses (Chao EC et al. Hum. Mutat. 2008 Jun;29:852-60). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
