This variant lies in the CFTR gene (transcript NM_000492.4) at 4 bases into the intron immediately after coding-DNA position 579, deleting one base. Submitter rationale: Variant summary: CFTR c.579+4delT alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 249988 control chromosomes. c.579+4delT has been reported in the literature in individuals affected with Cystic Fibrosis (Raraigh_2022, Schrijver_2016). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
