NM_000527.5(LDLR):c.1137T>A (p.Cys379Ter) was classified as Pathogenic for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C379* pathogenic mutation (also known as c.1137T>A), located in coding exon 8 of the LDLR gene, results from a T to A substitution at nucleotide position 1137. This changes the amino acid from a cysteine to a stop codon within coding exon 8. This variant has been observed in at least one individual with a personal and/or family history that is consistent with familial hypercholesterolemia (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
