This variant lies in the TNNC1 gene (transcript NM_003280.3) at coding-DNA position 334, where A is replaced by G; at the protein level this means replaces isoleucine at residue 112 with valine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with valine at codon 112 of the TNNC1 protein (p.Ile112Val). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and valine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
