Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004281.4(BAG3):c.1088A>C (p.Glu363Ala), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the BAG3 gene (transcript NM_004281.4) at coding-DNA position 1088, where A is replaced by C; at the protein level this means replaces glutamic acid at residue 363 with alanine — a missense variant. Submitter rationale: Variant summary: BAG3 c.1088A>C (p.Glu363Ala) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251484 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1088A>C in individuals affected with Myofibrillar Myopathy, BAG3-Related and no experimental evidence demonstrating its impact on protein function have been reported. Three submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
