Uncertain significance for SHORT syndrome; Immunodeficiency 36 with lymphoproliferation; Agammaglobulinemia 7, autosomal recessive — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_181523.3(PIK3R1):c.346C>A (p.Pro116Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with threonine at codon 116 of the PIK3R1 protein (p.Pro116Thr). The proline residue is highly conserved and there is a small physicochemical difference between proline and threonine. This variant is present in population databases (rs367923318, ExAC 0.001%). This variant has not been reported in the literature in individuals with PIK3R1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
