Pathogenic for X-linked myopathy with postural muscle atrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001159699.2(FHL1):c.505T>C (p.Cys169Arg), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Cys153 amino acid residue in FHL1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 23965743). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 11552). This missense change has been observed in individual(s) with clinical features of X-linked dominant FHL1-related disorders (PMID: 18274675; Invitae). In at least one individual the variant was observed to be de novo. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 153 of the FHL1 protein (p.Cys153Arg).
