Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.5365-5A>G, citing Ambry Variant Classification Scheme 2023: The c.5365-5A>G intronic variant results from an A to G substitution 5 nucleotides upstream from coding exon 24 in the DICER1 gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice acceptor site; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear.
