Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_181486.4(TBX5):c.125C>T (p.Pro42Leu), citing Ambry Variant Classification Scheme 2023: The p.P42L variant (also known as c.125C>T), located in coding exon 1 of the TBX5 gene, results from a C to T substitution at nucleotide position 125. The proline at codon 42 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
