The aspartic acid at codon 375 is replaced by glycine, an amino acid with similar properties. Based on data from gnomAD, the G allele has an overall frequency of 0.0049% (10/203330) total alleles studied, with 2 hemizygote(s) observed. The highest observed frequency was 0.0357%% (10/27989) of Latino alleles. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
