NM_003072.5(SMARCA4):c.2506-5T>C was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2506-5T>C intronic variant results from a T to C substitution 5 nucleotides upstream from coding exon 17 in the SMARCA4 gene. This variant was detected as heterozygous in individual(s) with no reported features of Coffin-Siris syndrome (Ambry internal data). This nucleotide position is poorly conserved in available vertebrate species. In silico splice site analysis for this alteration is inconclusive. RNA evidence is insufficient at this time (Ambry internal data). Based on the supporting evidence, the association of this alteration with rhabdoid tumor predisposition syndrome is unknown; however, the association of this alteration with Coffin-Siris syndrome is unlikely.
