This variant lies in the PANK2 gene (transcript NM_001386393.1) at coding-DNA position 67, where A is replaced by G; at the protein level this means replaces methionine at residue 23 with valine — a missense variant. Submitter rationale: Variant summary: PANK2 c.397A>G (p.Met133Val) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 68142 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.397A>G has been reported in the literature in the compound heterozygous state in at least one individual with clinical features of Pantothenate Kinase-Associated Neurodegeneration (e.g. Zhai_2021). This report does not provide unequivocal conclusions about association of the variant with Pantothenate Kinase-Associated Neurodegeneration. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
