Pathogenic for Hereditary pheochromocytoma and paraganglioma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_017849.4(TMEM127):c.245-1G>T, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects an acceptor splice site in intron 2 of the TMEM127 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with pheochromocytomas (PMID: 20154675). This variant is also known as IVS2-1G>T. ClinVar contains an entry for this variant (Variation ID: 109). Studies have shown that disruption of this splice site results in activation of a cryptic splice site , and produces a non-functional protein and/or introduces a premature termination codon (internal data).
