Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 177615 control chromosomes. c.310C>T has been observed in individuals affected with Nephrogenic Diabetes Insipidus (e.g. Inaba_2001, Miyakoshi_2003, Mizuno_2004, Keenswijk_2017, Li_2021). These data indicate that the variant is likely to be associated with disease. At least two publications reports experimental evidence evaluating an impact on protein function and found that the variant results in reduced binding capacity compared to the WT protein (e.g. Inaba_2001, Faerch_2009). The following publications have been ascertained in the context of this evaluation (PMID: 19812297, 11232028, 27350623, 34101133, 14709855, 15249704).
