Pathogenic for Renal cyst; Polycystic kidney disease, adult type — the classification assigned by (GEEPAD) Grupo de Estudio de la Enfermedad Poliquística Autosómica Dominante, Hospitales Universitarios Virgen de las Nieves y San Cecilio (Granada) to NM_001009944.3(PKD1):c.10527_10528del (p.Glu3509fs), citing ACMG Guidelines, 2015. This variant lies in the PKD1 gene (transcript NM_001009944.3) at coding-DNA position 10527 through coding-DNA position 10528, deleting 2 bases; at the protein level this means shifts the reading frame starting at glutamic acid residue 3509, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: It is a frameshift-type nucleotide change in which GA nucleotides are lost at positions 10527 and 10528 of the coding sequence and which predicts an amino acid change from Glutamic to Aspartic at codon 3509, causing an alteration of the reading frame and appearance of a premature stop codon The variant has been identified in 10 of our patients from 6 unrelated families, all of them afected with renal cysts . The age range at diagnosis in our cases is between 8 and 62 years old. This variant has been described before by Yu C,et al. (BMC Med Gen.2011,12:164), although variant is not found in gnomAD exomes and in gnomAD genomes databases.
