Pathogenic for Hereditary spastic paraplegia 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_89595875)_(89595997_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in SPG7 are known to be pathogenic (PMID: 21623769, 22964162). A similar deletion of exon 6 has been reported to segregate with autosomal recessive hereditary spastic paraplegia in a family¬† (PMID: 23733235). This variant is an out-of-frame deletion of the genomic region encompassing exon 6 of the SPG7 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product.
