Submitter rationale: This sequence change results in a premature translational stop signal in the LAMA2 gene (p.Pro3053Glnfs*26). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 70 amino acid(s) of the LAMA2 protein. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with LAMA2-related conditions. This variant disrupts the C-terminus of the LAMA2 protein. Other variant(s) that disrupt this region (p.Lys3074Asnfs*5) have been determined to be pathogenic (PMID: 20207543).
