Pathogenic for Joubert syndrome; Meckel-Gruber syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_56296502)_(56296882_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in MKS1 are known to be pathogenic (PMID: 19466712, 24886560, 26490104). This variant has been observed in individual(s) with Bardet‚ÄìBiedl syndrome (PMID: 22353939, 29620724). This variant is a gross deletion of the genomic region encompassing exon 1 of the MKS1 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 1 of the MKS1 gene. This is expected to result in an absent or disrupted protein product.
