This variant disrupts a region of the CNGA1 protein in which other variant(s) (p.Arg629*) have been determined to be pathogenic (PMID: 24154662; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. ClinVar contains an entry for this variant (Variation ID: 1076607). This premature translational stop signal has been observed in individual(s) with retinitis pigmentosa (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ala525Leufs*14) in the CNGA1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 166 amino acid(s) of the CNGA1 protein.
