Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000008.10:g.(?_119936613)_(119964060_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the TNFRSF11B gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Whole-gene deletions of TNFRSF11B have been reported to be homozygous in individuals affected with juvenile Pagetâ€šÃ„Ã´s disease (PMID: 12124406). Loss-of-function variants in TNFRSF11B are known to be pathogenic (PMID: 9647741, 26762549). For these reasons, this variant has been classified as Pathogenic.
