Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.10:g.(?_107802294)_(107807156_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an in-frame deletion of the genomic region encompassing exon(s) 3-4 of the COL4A5 gene. It preserves the integrity of the reading frame. This variant has not been reported in the literature in individuals with COL4A5-related conditions. For these reasons, this variant has been classified as Pathogenic. This deletion contains glycine residues within the Gly-Xaa-Yaa repeats of the triple helix domain which are required for the structure and stability of fibrillar collagens (PMID: 7695699, 8218237, 19344236). In COL4A5, variants at these glycine residues are significantly enriched in individuals with disease (PMID: 23720012, 27627812) compared to the general population (ExAC).
